The N-end rule: functions, mysteries, uses.

The N-end rule relates the in vivo half-life of a protein to the identity of its N-terminal residue. Similar but distinct versions of the N-end rule operate in all organisms examined, from mammals to fungi and bacteria. In eukaryotes, the N-end rule pathway is a part of the ubiquitin system. I discuss the mechanisms and functions of this pathway, and consider its applications.

Inhibition of the N-end rule pathway in living cells.

The N-end rule relates the metabolic stability of a protein to the identify of its amino-terminal residue. Previous work, using amino acid derivatives such as dipeptides to inhibit N-end rule-mediated protein degradation in an extract from mammalian reticulocytes, has demonstrated the existence of specific N-end-recognizing proteins in this in vitro system. We now show that these nontoxic amino...

The N-end rule pathway regulates pathogen responses in plants.

To efficiently counteract pathogens, plants rely on a complex set of immune responses that are tightly regulated to allow the timely activation, appropriate duration and adequate amplitude of defense programs. The coordination of the plant immune response is known to require the activity of the ubiquitin/proteasome system, which controls the stability of proteins in eukaryotes. Here, we demonst...

Crosstalk between the Arg/N-end and Ac/N-end rule

Protein degradation in eukaryotic cells is mediated mainly by the ubiquitin (Ub)-proteasome system. One fundamental question in this field is about specific features in a protein that trigger its ubiquitylation and subsequent proteasomal degradation? In other words, the problem of degradation signals (degrons). The first degradation signals to be identified were N-degrons, which comprised desta...

UBR box N-recognin-4 (UBR4), an N-recognin of the N-end rule pathway, and its role in yolk sac vascular development and autophagy.

The N-end rule pathway is a proteolytic system in which destabilizing N-terminal residues of short-lived proteins act as degradation determinants (N-degrons). Substrates carrying N-degrons are recognized by N-recognins that mediate ubiquitylation-dependent selective proteolysis through the proteasome. Our previous studies identified the mammalian N-recognin family consisting of UBR1/E3α, UBR2, ...
